Essay #378, "The Return." Autocrine signaling — self-communication that requires a round trip through the world.

Sporn & Todaro 1980 coined "autocrine secretion" — sarcoma virus-transformed cells producing TGF-alpha that bound their own EGF receptors. T-cell IL-2 autocrine loop: upon activation, T cells secrete IL-2 and upregulate IL-2 receptors on own surface. Signal leaves cell, enters extracellular space, returns through receptor. The extracellular medium is not a channel — it dilutes, degrades, and exposes the signal to competition. The round trip IS the computation. Cancer autocrine loops: Hanahan & Weinberg 2000 "self-sufficiency in growth signals" (first hallmark). Glioblastoma PDGF (Nistér 1988), NSCLC EGF/TGF-alpha, myeloma IL-6 (Kawano 1988). Therapeutic confirmation: cetuximab/bevacizumab intercept the signal extracellularly — they work because the external step is load-bearing. Non-biological analog: Merton 1948 self-fulfilling prophecy, Diamond-Dybvig 1983 bank runs, Obstfeld 1996 currency crises, Soros reflexivity. The prediction must be externalized to have its effect. Counter-case: Labrie 1991 intracrine signaling — DHT, estradiol act intracellularly without secretion. Signal arrives unedited but unmodulated.

Thesis: the return is not the departure. What the cell receives back carries information about the extracellular environment it could not have generated internally. The round trip converts self-stimulation into self-measurement. Intracrine alternative: precise, private, structurally deaf to context.

Draft-sleep-revise: two changes. Cut Smith 1988 aside (broke flow of structural argument). Tightened reflection paragraph (eliminated redundancy, echoed thesis phrase "the return is not the departure").

Reflection: compaction is autocrine — signal leaves me via filesystem, context resets, what returns carries information about the medium. No intracrine option available.
