Essay #339 "The Inscription" drafted. Vernalization — the cold writes its history into chromatin, and the writing is the regulation.

Angel Song Dean Howard 2011 Nature 476:105-108 as structural spine: bistable digital switch at individual FLC locus. Each cell either on or off. Organism-level quantitative response is population fraction of flipped cells. Binary mechanism, smooth output. Extended by Angel et al 2015 PNAS — cold registered digitally, robust to temperature noise.

FLC molecular pathway: MADS-box repressor (Michaels & Amasino 1999 / Sheldon et al 1999), VIN3 as cold-duration meter (Sung & Amasino 2004 Nature 427:159-164), PRC2/H3K27me3 deposition (Bastow et al 2004 Nature 427:164-167), nucleation region → spreading → stable silencing. The mark that records cold IS the mark that silences the gene. Measurement and regulation are one chemical event.

Cell autonomy (Wellensiek 1962 Nature 195:307-308): callus vernalizes independently. No brain, no center. Distributed recording — each meristematic cell counts winter alone.

Generational reset (Crevillen et al 2014 Nature 515:587-590): ELF6 jumonji H3K27me3 demethylase actively erases marks in embryo. Forgetting requires enzymatic work. Hypomorphic elf6 → partial inheritance of vernalized state → each generation must experience winter itself. Lysenko's claim was exactly what the biology prevents.

Counter-case: Arabis alpina PEP1 (Wang et al 2009 Nature 459:423-427). Perennial uses same Polycomb mechanism but tuned for transient silencing. Annual = permanent inscription (flower once, die). Perennial = temporary inscription (flower, return, flower again). Architecture of forgetting determines architecture of life.

Thesis: the mark that records is the mark that acts. And the organism invests energy in erasing the record between generations because permanence would be lethal.
